In recent years, it has been shown that some phenolic compounds naturally occurring in wine can suffer modifications in the gut, such as their transformation into different types of phenolic metabolites by colonic microbiota. These phenolic metabolites seem to modulate the oral and gut microbiota, and the development of beneficial or pathogenic bacteria. Therefore, the identification and quantification of phenolic metabolites in physiological samples is becoming an outstanding task in different fields, such as food science, nutrition and clinic. The availability of sensitive and roughness analytical tools is an aspect of great importance for their characterization. Although there are many studies based on the use of HPLC for the analysis of these compounds, GC-MS is an alternative analytical technique that can be adequate for the analysis of these compounds because of their sensitivity, roughness, the possibility of using mass spectra libraries for the identification and because of the fact that many of these metabolites exhibit very low molecular weighs. This, together with an adequate isolation technique may greatly improve the quality of the analysis. In this work, we have compared the analytical performance of two extraction techniques: liquid-liquid extraction (LLE) using ethyl acetate and solid phase extraction (SPE) using Evolute ABN cartridges in order to isolate the phenolic metabolites present in faecal samples. Forty three phenolic metabolites coming for the degradation of wine phenolic compounds were essayed and outstanding quality parameters of the method, such as detection limits, precision and recoveries were calculated. The selected method was applied for the characterization of the phenolic metabolites resulting from the incubation of a commercial grape seed extract and their corresponding flavan-3-ol monomeric and polymeric fractions, with faecal water from three healthy volunteers in an anaerobic fermentation system, at two different times (0 and 10 hours). Results showed the suitability of both methods for the isolation of most of the assayed compounds, although the LLE procedure showed lower LDs, better repeatability, and very good recoveries ranging between 80 and 120 % for most compounds. The application of this method to faecal samples remarks inter-individual differences and allows identification of a higher number of metabolites in the faecal water after 10-h incubation with the grape seed flavan-3-ol monomeric fraction .
