6-Benzylaminopurine (BAP) concentration on shoot proliferation of 'Gala' apple in vitro
- Seven different concentrations of BAP (6-benzylaminopurine) growth regulator were tested to maximize in vitro proliferation of 'Gala' apple tree explants. The basic culture medium consisted of the Murashige & Skoog saline solution, 10.0 mg/liter of thiamine, 1.8 mg/liter of micotine acid, 2.5 mg/liter of pyridoxine, 100 mg/liter of inoxitol, 80 mg/liter of cysteine, 6.5 g/liter of agar-agar, 30 g/liter of sucrose, supplemented with BAP at 6.0, 7.0, 8.0, 9.0, 10.0, 11.0 and 12 mm concentrations. Significant micrease in the number of viable shoots was obtained in the 10.0 mm of BAP concentration, ca. 16.1 propagules per explant. The "vitrification" level had and increase of 5% for 10 mm and up to 40% for 12.0 mm of BAP, reducing considerably the propagules useful for the rooting phase. The quadratic function fitting was of 83%, showing a maximum of sprouting at the 9.3 mm of BAP concentration
