Polymorphism of the PrP gene in Czech sheep breeds

Variant forms of the prion protein PrP are associated with natural scrapie in different sheep breeds. Breeding programmes thus may be developed as one of possible measures for the control of scrapie. For that purpose, information on PrP polymorphisms and resistance to scrapie in local breeds is required. A pair of primers amplifying a 227 bp long PCR product containing codons 136,154, and 171 was designed. Polymorphism of the region amplified was analyzed by PCR-RFLP and PCR-SSCP techniques. As reference breeds, Suffolk and Texel were analyzed. Different haplotypes were detected and inter-breed differences were observed as expected. The results will be compared with results of PCR-SSCP and sequence analysis in Czech breeds that this currently underway. (Texte intégral)
