Ploidy levels in native and cultivated plants of the Amaryllidaceae family from Paraguay

The Hippeastreae tribe is taxonomically complex, karyologically variable, and phytochemically and ornamentally important, and its center of distribution is located in subtropical South America. Little cytological and reproductive information on this tribe is available on native material from Paraguay. In order to contribute to the characterization of native Paraguayan plant resources of this tribe, the ploidy level has been determined using cytogenetic techniques, flow cytometry, and fertility reproductive characterization. Individuals from natural populations and cultivated materials from the Central Department, were analyzed during 2019-2022. The following chromosome numbers were found: Habranthus brachyandrus (Baker) Sealy, 2n = 4x =24; Habranthus tubispathus (L’Hér.) Traub, 2n = 4x = 24; Hippeastrum puniceum (Lam.) Voss, 2n = 2x = 22; H. vittatum (L’Her.) Herb., 2n = 4x = 44; and from Z. mesochloa Herb. ex Lindl., 2n = 2x = 12. By histo-foliar flow cytometry the diploid ploidy level of individuals of Hippeastrum reticulatum (L’Hér.) Herb and tetraploid of Habranthus brachyandrus were confirmed. Fertility analyzed as pollen viability and mean seed production per capsule was 79% and 20 seeds/ capsule in Hippeastrum puniceum, 96% and 12 seeds/capsule in H. vittatum, 83% and 43.75 seeds/ capsule in Z. mesochloa, and 65.25 seeds/capsule in H. brachyandrus. The contributions on fertility, chromosome numbers and ploidy level of Habranthus brachyandrus, H. tubispathus, Hippeastrum vittatum and H. reticulatum are new records for Paraguay.
