Isolation of the cyanobacterium Leptolyngbya sp. growing in batch cultures and quality analysis of its biomass.
The cyanobacterium Leptolyngbya sp. It is characterized by being a source of enzymes, toxins, auxins, and pigments, which is why further studies are required due to its biotechnological potential. The objective of this work was to characterize a strain of the cyanobacteria Leptolyngbya sp., using fed-batch cultures, and to assess the biochemical quality of the biomass and analyze its possible toxicity in zebrafish. For this, samples were taken in the “Sixto Durán Ballén” reservoir, Manabí, Ecuador and for their isolation a combination of techniques was used by streaking on agar plates and serial dilutions in BG110 medium. Subsequently, the cultures were scaled from 10 mL to 14 L. The cultures were maintained at different light intensities of 2, 4 and 8 Klux; with a photoperiod of 24:0, at 20 ⁰C ± 2 and BG110 medium enriched with 1 and 5 mL of NaNO3 (0,085 and 0,425 mM; respectively). Chlorophyll and phycocyanin variables were monitored during the experiments. A proximate analysis of the harvested biomass and acute toxicity of the culture supernatant and the lipid fraction of the biomass were carried out, using zebrafish (Danio rerio) as a test organism. 5,06±0,15 mL of biomass/10 mL of culture was obtained, dry biomass of 1,5 g/L, with a chlorophyll a content of 9,04±0,76 µg/L and phycocyanin of 0,013± 0,003 mg*L-1 Likewise, 29,59, 25,50 and 27,49 % of carbohydrates, proteins, and ashes; respectively. Toxicity is reported in the supernatant and fat-soluble fraction at 100, 50 and 25 %, and null at 10% and in controls. This Leptolyngbya strain showed growth capacity, in batch cultures, high production of proteins and phycocyanin, at low concentrations of NaNO3 in BG11o medium and expressed toxicity on zebrafish. Monitoring is recommended when its presence is observed in lentic water bodies.
