In vitro GAS PRODUCTION AND FERMENTATIVE CHARACTERISTICS OF RUMINAL CELLULOLYTIC BACTERIAL CONSORTIA OF WATER BUFFALO (Bubalus bubalis) AND SUIZ-BU COW.

The cellulolytic bacterial consortia (CBC) serve as additives in the feed of ruminants to improve the degradation of cellulose and hemicellulose. The objective of the present study was to determine the in vitro production of gas and the fermentative characteristics of CBC of water buffalo and Suiz-bu cow in coculture with ruminal bacteria of Suiz-bu cow. The ruminal fluid of a water buffalo and of a Suiz-bu cow were inoculated in media of crystalline cellulose Whatman® paper to obtain CBC of the buffalo cow (CBCbuf) and of the Suiz-bu cow (CBCcebu). The experimental design was completely randomized and the treatments were the inocula: total ruminal bacteria (TRB), CBCceb, CBCbuf, cocultureceb (TRB and CBCceb) and coculturebuf (TRB and CBCbuf). The biodigesters contained 0.5 g of cobra grass, 45 mL of culture medium and 5 mL of inoculum. Gas production was measured at 3, 6, 9, 12, 24, 48 and 72 h, and that of methane (CH4 ) at 24, 48 and 72 h. The variables evaluated were pH, ammonia nitrogen (N-NH3 ), dry matter degradation (DEGDM) and NDF degradation (DEGNDF), and the bacteria were counted. CBCbuf and CBCbov produced a higher amount (p£0.05) of gas and CH4 accumulated at 72 h, and increased (p£0.05) DEGDM and DEGNDF, but without differences between them (p>0.05). TRB did not present differences (p>0.05) with the cocultureceb in the DEGDM and with coculturebuf in the DEGNDF. The inocula with CBC did not present differences (p>0.05) in pH. Furthermore, the bacteria count and N-NH3 were similar in the media evaluated with different source of inoculum (p>0.05). The cellulolytic bacterial consortia increased production of gas, CH4 , DM degradation and NDF degradation when they were added to the ruminal bacteria under in vitro conditions.
