Identification of novel loci underlying tolerance to Clavibacter michiganensis originating from Solanum arcanum LA2157

In the study, we used an advanced BC3S6 line (and its selfing) originating from a cross between S. arcanum LA2157 and the tomato cv. Moneymaker. Plants in the line were inoculated with Clavibacter michiganensis strain NCPP382. Based on symptom development plants were categorized as susceptible (S) or resistant/tolerant (R). Whole-genome sequencing (WGS) of a susceptible and a resistant pool of 14 plants each was done to identify loci involved in the observed phenotypes. Genomic DNA of plants was extracted from leaves. For each bulk, 28.57 ng of gDNA of each individual were pooled. For the WGS , 400 ng of (pooled) gDNA were used for the library preparation and sequencing. Sequencing was done on an Illumina Novaseq 6000 platform producing 151 bp paired end reads at a 35x depth (Novogene Europe, Cambridge, United Kingdom).
