Determinación de enzimas de resistencia en tribolium castaneum (herbst) (coleoptera: tenebrionidae)

ABSTRACT: This research work was carried out at “Universidad Autónoma Agraria Antonio Narro” in Saltillo, State of Coahuila, Mexico, in the agricultural Parasitology Department, from January 2008 to December 2009. The purpose of this work was to determinate by means several diagnosis methods, the susceptibility and biochemical resistance mechanism; as well as the intraspecific variability of T. castaneum obtained of flour. Flour beetles were collected on Saltillo, Nuevo Leon and Aguascalientes. Flour beetles were transfer to laboratory of toxicology of Universidad Autonoma Agraria Antonio Narro to determinate the level of resistance and enzymatic mechanisms of resistance. The first method was to determinate the susceptibility and present mechanism was bioassay, by means of the residual film technique, evaluating populations of T. castaneum against insecticides malathion, bifentrine endosulfan and cipermetrine. The second method used was based on a series of biochemical tests (microplate assay), microplate methods had to be adapted from the methods already described for mosquitoes (Brogdon et al, 1983, 1984, 1988, 1990 and 1997), to determinate the levels of α-esterase, β-esterase, oxidase, glutahtione s-transferase and acetyl cholinesterase.
