In vitro assessment of the fertilizing capacity of frozen-thawed semen from Rubia Gallega bulls

With the purpose of assessing the discriminative power of the tests for the evaluation of the fertilization capacity of post-thaw semen, the relationship between the routine tests sperm-gram that are used for evaluating semen samples in vitro and some complementary tests that evaluate the functional capacity of the spermatozoa were studied. As routine sperm-gram tests, progressive post-thaw motility (%MOV), live spermatozoa (%EVIT) and the incidence of normal sperm (%ENOR and %GNOR) were considered. As complementary tests, the hypo-osmotic swelling test (%ENDO), acrosom integrity (%GNAR), motility (%MOVS) and sperm concentration (CONS) after swim-up, rate of penetrated oocytes (%P), divided embryos (%D) and the development of blastocyts (%B) after 7-9d of cultured in vitro were examined. Frozen-thawed semen from 7 Rubia Gallega bulls (8 tubes of the same ejaculate) were evaluated. The frozen-thawed spermatozoa of 3 tubes were subjected to a swim-up procedure and those separated in this way were tested for in vitro fertilization (FIV) on 2,567 oocytes. There were significant differences (P<0,05) between bulls for all sperm traits. No significant relation between the routine sperm-gram tests and the FIV were found. Positive significant correlations between %ENOR and %GNOR, between %EVIT and %ENDO, %ENOR, %MOV and CONS, between %GNAR and %MOV, %P, %D, %MOVS and CONS, between %P and %D, and between %ENDO and %ENOR, %P, %D, %MOVS and CONS were found. The complementary tests along with the routine sperm-gram allowed the classificaevaluation sperm-gram, in order to more accurately assess the fertilizing capacity of a semen sample.
