Molecular analysis of the procine AR gene as a candidate for health and reproduction

The androgen receptor (AR) a mediates androgenic actions by regulating transcription of target genes which involve many processes of development and maintenance of sexual dimorphism along life-span. Ubiquitous expression of the AR in diverse tissues and cell types especially of reproductive and immune systems suggests its important direct roles for traits related to these tissues. Many evidences obviously show that mutations of the AR associate with various disorders in human and animals. Recently, QTLs for backfat thickness and intramuscular fat content were positional mapped at the AR locus. Therefore AR is an interesting candidate gene for pig health, reproduction, and also performance. 'Me aims of this study were isolation of the porcine AR and detection of polymorphisms as a tool for association studies and analysis of functional properties of the porcine AR. The complete cDNA sequence of the porcine AR gene was obtained starting with heterologous primers designed from conserved regions of the human and rat AR to obtain the sequence of the central region of the porcine gene and subsequent 5'and 3'-RACE using homologous primers (AccNo: AF161717). Sequencing of the gene promoter sequence of 2,459 bp was achieved by means of PCR screening of a lamda phage DNA library (AccNo: AF331845). Comparative sequencing of five pig breeds revealed eight SNPs and two INDELs in the promoter region. Three SNPs in 5'-UTP, one microsatellite (CCTTT)n in intron 1 of 5'-UTR, and a CAG INDEL in exon 1 were identified. These polymorphisms are segregating in the DUMI F2 resource population and two different haplotypes are found, one inherited from Duroc and the other from Berlin Miniature Pig grandparents. AR mRNA expression in 10 adult pig tissues was determined by real-time RT-PCR and showed the highest AR mRNA expression in ovary (100%). The relative AR gene expression to ovary were 83.9, 61.6, and 47.4% in adrenal gland, uterus, and liver, respectively. The transcription rate in pituitary, tonsil, muscle, and mammary gland was low (~ 1%) and even lower in leukocyte and jejunum. Differential gene expression in liver of the two haplotypes was investigated in eight litter mate adult pigs. Hemizygous male pigs carrying the AR haplotype from the Berlin-Miniature Pig showed the higher relative AR expression to beta-actin transcript than did those with the Duroc haplotype (P= 0.054). It can be concluded that the porcine AR gene is a highly polymorphic and differentially regulated gene and a potential candidate gene for traits related to pig health, reproduction, and performance.
