Integration of AFLP markers into an RFLP linkage map of sorghum

The AFLP technique has been applied in establishing an extended linkage map of sorghum. Twelve couples of primers yielded in total, 1380 bands out of which 181 were polymorphic in a 110 recombinant inbred lines population. Finally the AFLP linkage map of 892 cM consists of 147 loci distributed in 11 linkage groups, 34 markers remained independent. The AFLP loci were evenly distributed over the 11 linkage groups except the linkage groupe where the number of AFLPs is lower. The integration of AFLPs into an existing linkage map based on RFLP markers led to: - an increase of the length map (1886 cM versus 1350 cM for the RFLP map); - no disturbance in the original order of the RFLP markers. However, the order of AFLPs within the mixed linkage groups was slightly different; - in most cases, AFLP clusters filled in regions poorly marked or corresponding to earlier gaps. The results are discussed with possible applications of AFLP markers for accelerating a QTL-analysis and marker-assisted backcrossing. (Texte intégral)
