Mitochondrial DNA in fish taxonomy
Ethanol preserved tissues were digested in protease and mitochondial DNA (mtDNA) isolated through phenol, chloroform and ethanol extractions. Using Polymerase Chain Reaction (PCR) amplified double strand mtDNA (ds-mtDNA) was cut using restriction enzymes and the mtDNA fragments were subjected to electrophoresis. The migration of mtDNA bands in agarose wasobserved. The amplified single strand mtDNA (ss-mtDNA) was sequenced for base pairs. The order of this base pairs in different species was compared
