Optimización de una metodología para la inducción y proliferación de embriones somáticos de yuca (Manihot esculenta Crantz). Tesis (Ingeniero Agrónomo)

Somatic embryogenesis is the only method of cassava regeneration and it is actually used to obtain transgenic plants. Because of the low percentile of embryos that regenerate to plants, a great amount of embryogenic material for transformation must be needed. In our experiments, using MCOL1505 cultivar, various factors involved in primary and secondary somatic induction, and embryo proliferation were evaluated. The results suggested an optimal embryogenic efficiency with 4 and 8 mg/1 by 2, 4-D and Picloram; differences between these hormones were founded only during primary embryos induction. If the media is suplemented with 1 to 8 RM of cooper sulfate, the process could the improved. The use of casein hidrolizate didn't affect the primary embryos induction and proliferation, in contrast with secundary embryos induction, where the lowest response is observed, when cassein hidrolizate was used at the highest levels; the friable callus didn't respond to casein concentration. Also, the explant type was very important: in vitro inmature leaves and greenhouse meristems were the best explants. In the proliferation phase, the high density embriogenic groups and the fragmentation treatment, increased the embryo production and decreased the friable callus amount. When the cotiledonary and not elongated embryos were divided, the cotiledonary leaves were the best explants to secundary embryo induction. The germinated embryo withouth cotiledonary leaves were transfered to elongation phase, showing no negative effect on shoot elongation. Complementarilly, the proliferation control media (solid) was compared with liquid media, and it seems to produce an increase in embryo proliferation, and less friable callus production; in the germination phase, the liquid media proliferated embryos produced lower germinated embryos than solid media
