Contaminaçao fúngica, bacteriana e oxidaçao in vitro de explantes de Coffea arabica L. cv. catuaí LCH-2077-2-5-44

The contamination are a very large problem for tissues culture works, especially when the explants area extracted from plants maintained in the field. The subject of this study was to evaluate the contamination by bacteria and fungi and oxidation of roots. Leaves and stems segments of Coffea arabica L. cv. Catuaí line CH 20-72-2-5-44 inoculated of MURASHIGE & SKOOG (1962) "in vitro". The treatments were Na(0)C1 at 0.0, 5000, 10000, 20000 and 40000 ppm concentrations. The explants were cultured on "MS" medium supplemented with clorophenicol-5.5.mg/l, ascorbic acid-100 mg/l and tween 20-2 drops. The contamination by bacteria was not affected by Na(0)Cl. The contamination and the oxidation have a independent behaviour. The lowest contamination by fungi were observed up 20000 ppm Na(0)Cl for leaves explants, up 5000 ppm for roots explants and 40000 ppm for stems explants. The oxidation in leaves explants was induced by contamination and Na(0)Cl high concentrations.
