Infection of cacao seedlings by Moniliophthora roreri

Seeds of Theobroma cacao (cv Pound-7) and T. bicolor free of seeds coat and tegment were maintained for 3 days over blotter paper in a humid chamber. The unwounded seeds were inoculated by atomizing spore suspension of Moniliophthora roreri (10 exponent 5 conidia/ml added with 0.01 percent Tween 80), incubated for 48 hr in humid chamber under greenhouse conditions (22-26 grade centigrade) and planted in plastic pots containing sterilized soil. Eight wk after inoculation, T. bicolor seedlings expressed light brown necrosis at cotyledonary node, persisting mummified cotylendons attached, whereas in cacao seedlings these symptoms appeared 12 wk after inoculations. The T. bicolor seedlings presented wilting and death of seedlings 90 days after inoculation and cacao seedlings showed constriction at necrotic area but maintained normal growth. M. roreri was re-isolated from tissues of infected seedlings of T. cacao and T. bicolor.
