The incorporation of long-chain polyunsaturated fatty acids into infant formulas in the last years has contributed to enhance their susceptibility to lipid oxidation. Evaluation of lipid oxidation in infant formulas is usually approached by determination of the peroxidevalue (PV) of the lipid fraction extracted with organic solvents. However, due to the lack of specificity of the PV, anomalous results can be obtained when minor substances other than lipids are present in the extract. In this study, the PV was assessed by using lipid extracts obtained from commercial infant formulas with different organic solvents. The total lipids and the free and encapsulated lipid fractions were analysed. The free fraction was quantitatively extracted with hexane, while the total lipids as well as the encapsulated fraction were qualitatively extracted applying two methods. One of the methods consisted of a modification of the method of Folch-Less, that is, of a single extraction with a mixture of dichloromethane:methanol (2:1, v/v). The other one consisted of three extractions with hexane after disruption of the solid matrix with a mortar. The PV was compared with the quantitative determination of the polar compounds by the combination of adsorption and high-performance size-exclusion chromatography. Both the total lipids and the encapsulated lipids extracted with dichloromethane:methanol showed PVs significantly lower than those of the corresponding extracts obtained with hexane in the mortar method, or even equal to zero in various samples. Nevertheless, no significant difference in the content of total oxidation compounds was found. In view of likely interferences in the PV, the extracts were dissolved in hexane and treated with a single wash with water. Whereas the PVs of the extracts obtained with hexane in the mortar method were unchanged, those of the extracts obtained with dichloromethane:methanol increased after the wash with water and were close to those of the extracts obtained with hexane. These results suggested that substances of polar nature were present in the extracts obtained with the modified method of Folch-Less and interfered with the PV determination.
