Somatic embryogenesis of cocoa from floral parts.
Somatic embryogenesis from floral buds was tested on 25 cocoa genotypes under defined culture conditions, revealing genotypic variations. Histological monitoring of somatic embryo ontogenesis revealed that the somatic embryos were of multicellular origin. The right conditions for the selection and maintenance of meristematic cells (from which somatic embryos are derived) and embryogenic cells were sought by culturing in a liquid medium using a temporary immersion system (TIS). A friable embryogeniccallus was obtained. A study of zygotic embryogenesis was carried out with a view to improving the later stages of somatic embryogenesis. Zygotic embryogenesis was characterized by a period of embryo growth, followed by accumulation of starch and proteinreserves, during which slow and moderate desiccation occurred. By adding a growth phase to the somatic embryogenesis protocol, and defining a maturation medium containing abscisic acid, embryo maturation was improved.
