Cryopreservation de of shoots and seeds of cedar (Cedrela odorata L.)
In order to protect the forest resources of Cedrela odorata L., currently threatened, it was considered necessary to establish a protocol for its cryopreservation. Research was conducted using shoots and seeds. Vitrification techniques were evaluated with shoots, using various solutions and rapid freezing in liquid nitrogen (LN). Initially the best results were obtained using incubation for 5 min in PVS3 solution before freezing, survival being 61% after 3 weeks of culture on Murashige and Skoog (MS) medium with 1.0 mg.l-1 BAP and 0.1 mg.l-1 GA3. Moreover, by incubating the apices in 0.3 M sucrose solution for 5 min prior to freezing, the survival rate reached 86% after 2 weeks of culture on MS medium with 1.0 mg.l-1 BAP, 0.1 mg.l-1 GA3 and 0.01 mg.l-1 IAA. When incubation in the solution of 0.3 M sucrose was used and recovery MS medium with 1 mg.l-1 KIN, 0.5 mg.l-1 GA3 and 100 mg.l-1 activated charcoal, the appearance of the surviving shoots improved significantly. For cryopreservation of cedar seeds, the methodology of dehydration and rapid freezing in liquid nitrogen (LN) was evaluated. Seeds with moisture content between 7.9% and 5.5% were placed in 5 ml polypropylene vials and these were directly immersed in NL. The effect of the storage period in NL (1 h, 24 h, 7 days, 1 month, 3 months and 6 months) on seed germination was evaluated. There were no statistical differences in ability of seeds to germinate after storage during different periods.

En busca de proteger el recurso forestal de Cedrela odorata L., actualmente amenazado, se planteó la necesidad de establecer un proto- colo para su crioconservación; la investigación se realizó con ápices y semillas. Con los ápices se evaluó la técnica de vitrificación a través de varias soluciones vitrificadoras y congelamiento rápido en nitrógeno líquido (NL). Inicialmente los mejores resultados se obtuvieron al evaluar la incubación en la solución PVS3 por 5 min antes del congelamiento, con una sobrevivencia del 61% después de 3 semanas de cultivo en el medio Murashige y Skoog (MS) con 1,0 mg.l-1 BAP y 0,1 mg.l-1 AG3. Por otra parte, al incubar los ápices en una solución 0,3 M de sacarosa por 5 min previo al congelamiento, el porcentaje de sobrevivencia alcanzó 86% después de 2 semanas de cultivo en un medio MS con 1,0 mg.l-1 BAP, 0,1 mg.l-1 AG3 y 0,01 mg.l-1 AIA. Cuando se utili- zó la incubación en la solución de 0,3 M sacarosa y el medio de recuperación MS con 1 mg.l-1 KIN, 0,5 mg.l-1 AG3 y 100 mg.l-1 carbón activado, la apariencia de los ápices sobrevivientes mejoró considerablemente. Para la criopreservación de semillas de cedro, se evaluó el método de des- hidratación y congelamiento rápido en nitrógeno líquido (NL). Semillas con un contenido de humedad entre 7,9% y 5,5% fueron colocadas en criotubos de polipropileno de 5 ml y los criotubos sumergidos directamente en NL. Se evaluó el efecto del periodo de almacenamiento en NL (1 h, 24 h, 7 días, 1 mes, 3 meses y 6 meses) sobre la germinación de las semillas. No se presentaron diferencias significativas en su capacidad de ger- minación después de permanecer en los diferen- tes periodos de almacenamiento.
