Infected plants do not show any symptom, but contain toxíc alkaloids. Sínce some endophyte specíes are very similar morphologically, identification of these fungi often involve tedious laboratory work such as nucleotide sequencíng of variable genes, a process which involves multiple steps and is expensive. We have studied the potential of near infrared reflectance spectroscopy (NIRS) to classífy two species of the endophytic genus Epichloe. Epichloe festucae infects red fescue (Festuca rubra) and other fine fescues, whíle E. typhina infects several grass specíes. For this study we isolated E. festucae from plants of red fescue and E. typhína from orchardgrass (Dactylis glomerata). Pure cultures of E. fes tuca e and E. typhina are virtually identical, in terms of morphology and radial growth rates. Without information about the host precedence of the fungus, the best way of identifying them is by analysis of partial sequences of ribosomal RNA genes. For this work, the NIR spectra of 189 samples derived from 25 isolates of E. festucae and 17 ot E. typhina were registered. Two thirds of the isolates of each species were used to develop a calibration by means of principal component analysis. The remaining samples were used tor validation. With the calibration obtaíned, both endophyte specíes could be clearly separated, each specíes being in a different cluster. With the absorbance data, in 93% of the trials the test set was well classified. and the first component explained 98.5% of the variance.
