Vitellogenesis in the adult female of Blattella germanica requires a production of juvenile hormone (JH) much higher than that required for the nymphs to inhibit metamorphosis. In this respect, an interesting question is how the JH-producing glands, that is the corpora allata (CA), can cope with this hormonal hyperproduction. In females, CA biosynthetic activity decreases to very low values one day after the ecdysis to the last nymphal instar, and one day later, CA cells underwent cell hyperproliferation. We wonder whether there is a cause-effect relationship between both events, which can afford an answer to the above question. Here, we analyze the proliferation status of CA during the penultimate (fifth) and last (sixth) nymphal instar using EdU (5-ethynyl-2 ́-deoxyuridine), a thymidine analog that is incorporated to the DNA during its synthesis. Nymphs of different ages of the fifth and sixth instars were injected with 1 μl of 20 mM EdU solution, dissected three hours later and processed to fluorescence detection, based on a copper-catalyzed reaction between an azida (EdU) and a alkyne (Alexa Fluor® dye). Cell proliferation in the CA of fifth instar nymphs steadily increases from the fourth (N5D4) to the sixth day (N6D6). Recently molted sixth instar nymphs (N6D0) didn't show any proliferation in the CA, but just one day later the process starts again and reaches high levels in the second day (N6D2), which are maintained until the sixth day of development (N6D6). These proliferation rates are much higher than those observed in the fifth instar. Possible mechanisms by which a decrease of biosynthetic activity triggers cell hyperproliferation in CA cells in the last nymphal instar of B. germanica females will be discussed.
