Studies of a new lipid transfer protein (LTP) expresssed during coffee fruit development

The compounds present in coffee fruits directly determine the final quality of the coffee beverage. The identification of proteins expressed during coffee fruit development could contribute to understand key molecules linked to the final quality of coffee beans. Moreover, tissue-specific promoters are molecular tools that could be used to control the expression of genes improving coffee quality. This work presents data concerning expression analysis of a gene named CaLTP2 encoding a putative Lipid Transfer Protein (LTP) identified as preferentially expressed in coffee fruit within the Coffee ESTs Database. Northern blot analysis revealed that CaLTP2 is expressed only in fruits but not in roots and leaves, giving a transcript of approximately 600 b. In addition, CaLTP2 seems specific of endosperm (bean) because corresponding transcripts accumulate only during final stage of maturation but not in earlier developmental stages. To confirm this, CaLTP2 expression was also monitored by Real Time PCR (q-PCR) using RNA isolated from separated pericarp, perisperm and endosperm tissues. This showed that CaLTP2 was highly transcribed in endosperm, opening the way to the cloning and characterization of its corresponding promoter sequence that could be used in plant improving program.
