Multiplicación suspensiones celulares embriogénicas de Coffea arabica cv. Catimor 9722

The effect of inoculation density on the multiplication of embryonic cell suspensions of Coffea arabica cv. Catimor 9722 in a shaker, and the effects of two dissolved oxygen concentrations (30.0 and 80.0 percent) when the cell suspensions were cultivated in bioreactors were evaluated. Three inoculation densities (10.0, 25.0 and 50.0 gMF l-1) were evaluated in a shaker. Two bioreactors (model CMF-100, CHEMAP AG) were used for scaling up, both equipped with gas blending units for oxygen control and culture vessels with 2.0 litres capacity. It was shown that an inoculation density of 50.0 gMF l-1 produced the largest biomass and that subcultures should be carried out every 9 days when cell viability is still at 100 percent. It was observed that 80.0 percent of DO2 concentration markedly influenced the multiplication of cell suspensions and their composition producing the largest density (3.17 x 106 cells ml-1), and the best percentage of meristematic cells (90.85 percent) as compared with 30 percent DO2 and the control in shaker.
