Effect of abscisic acid (ABA) on the regulation of development of somatic embryos from Coffea canephora P. var. Robusta

The effect of ABA on regulation of embryogenesis in a liquid medium was investigated in C-R and M-229 clones of Coffea canephora P. sp. var. Robusta. 28-day-old calluses were subcultured in MIE and MME media, with 0, 0.5, 1.0 and 2.0 mg ABA L-1. When somatic embryos of different developmental stages were produced, Petri dishes containing MDE semisolid medium were inoculated. The total number of somatic embryos (SE), SE number per development stage and per clone were evaluated. Clone M-229 showed better development of SE. Embryo number was greatest with 1.0 mg ABA L-1 ABA concentration (1115), and was lowest with no ABA followed by 0.5 mg ABA L-1. The heart-shaped SE state reached values between 20.6 and 36.3 percent, while the figures for the globular state did not surpass 24.8 percent of embryos. The possibility of using ABA to diminish the degree of asynchrony in this crop was verified, as a result of its maturity effect on somatic embryos.
