Effect of BAP concentrations in pre-culture medium and BAP and TDZ in sub-culture medium on coffee micropropagation

The residual effects of BAP [benzyladenine] in a pre-culture medium and BAP and TDZ (thidiazuron) in a sub-culture medium on coffee axillary bud proliferation were evaluated. Nodal segments of coffee cv. Caturra Amarelo established in vitro were pre-cultured in MS medium without the growth regulators or with BAP at 6 mg/litre. After 180 days, the nodal segments were inoculated in MS medium supplemented with BAP at 0, 6, 9, 12 and 15 mg/litre, in the first experiment. In the second experiment, the nodal segments were inoculated in MS medium containing TDZ at 0, 0.05, 0.15, 0.45 and 1.35 mg/litre. The pH of the medium was adjusted to 5.8 and solidified with 0.7
