Distribuicao de Xylella fastidiosa no cafeeiro, nas regioes cafeeiras, e seu isolamento in vitro
Bacteria similar to Xylella fastidiosa were observed in xylem extracts of coffee plants (Coffea arabica) from several areas of Sao Paulo, from Capela do Alto, in the South, to Colina, in the North, and in Guaxupé, State of Minas Gerais. These extracts reacted positively in ELISA tests to antiserum to X. fastidiosa bacteria associated to Citrus Variegated Chlorosis (CVC), a sweet orange (Citrus sinensis) disease. As in oranges, the bacteria were detected in coffee roots, twigs, and leaves. Affected leaves usually showed symptoms of scalding. "Requeima do Cafeeiro" or "Coffee Leaf Scalding", therefore, is the suggested name for the probable coffee disease associated to X. fastidiosa. The coffee bacteria were isolated and cultured in PWS medium, which is used for the CVC bacteria, in early November, 1995, 15 to 20 days after inoculation of the medium with xylem extracts from veins and petioles of symptomatic leaves.
