Embryogènese somatique chez Coffea arabica induction et developpement des cellules embryogènes
In most of the cases studied up to now, the formation of somatic embryos has required culture of a given explant on two successive media, with different hormone balances: an induction medium on which embryogenous cells are initiated in a callus, and a differentiation medium on which the embryos can develop. Coffea arabica leaf explants follow this pattern, but they can also form embryos on a single medium containing either cytokinin alone or a mixture of cytokinin and auxin (direct embryogenesis). Histocytological study of the calluses obtained over a period of time on culture media that differ by their 2,4D concentration (from 0.1 to 1 mg/l) and the quality of the cytokinin (Kin. or BAP) has resulted in determining the optimum conditions for embryogenous cell induction, the best period for transferring them to the differentiation medium and the conditions for their subsequent development into embryos
