In an earlier work (Looser et. al., Phytochem., 13 (1974), 2515-2518), in which we fed potential precursors of caffeine together with L-methionine (methyl-14C) to leaf disks, we were able to determine the individual intermediates of a likely pathway of caffeine biosynthesis as far back as the nucleoside 7-methylxanthosine. We have now synthesized labelled 7-methylxanthosine (methyl-and ring-14C-lavelled) and have studied the 14C-incorporation into caffeine after feeding this tracer. The results confirm the proposed pathway from 7-methylxanthosine via 7-methylxanthine and theobromine to caffeine
