A technique for enhancing spore germination of Moniliophthora roreri

A technique was devised for obtaining consistently a high percentage germination of conidia of Moniliophthora roreri, agent of moniliasis of cacao. A suspension (10 exponent 6 conidia/ml) was prepared aseptically in sterile distilled water from an isolate grown for 15 days on potato carrot dextrose agar at 25 grade centigrade. Drops of this suspension were placed on prepoured (24 h) plates of Yeast Extract Agar (Oxoid Yeast extract 1 percent, Difco agar 1.5 percent) which were then incubated at lab. temperature (23-25 grade centigrade). Germination was assessed at intervals in samples stained with trypan blue or diluted cotton blue. Germination was observed after 4 h and c. 80 percent conidia germinated within 12 h, most by a single germ-tube. By 24 h mycelium had developed. Further tests showed that germination varied with age, isolate, pH and temperature.
