Despite the biogeographical and evolutionary interest of Socotra Archipelago as a part of the Gondwana supercontinent, very little is known about the origin, the tempo and the speciation mode its unique fauna (vicariance, colonization or intra-island diversification). The genus Haemodracon from Socotra include two species of endemic geckos, H. trachyrhinus and H. ribeckii, which are distributed in three different Socotran Islands (Socotra, Darsa and Samha). The two taxa are mostly found in sympatry and greatly differ in their behaviour, morphology and ecology. Yet we know little about their origin and evolutionary relationships, and in the particular case of H. trachyrhinus (Boulenger, 1899), it has never been included in any phylogenetic analysis. In the present study we try to shed some light in these issues, by assembling a dataset including 23 specimens of H. ribeckii (Peters, 1882) and 28 of H. trachyrhinus plus some representatives of its sister taxa from mainland Arabia, i.e. geckos of the genus Asaccus (Dixon & Anderson, 1973). All the specimens were sequenced for up to three mitochondrial (cytochrome b, 12S rRNA and 16S rRNA) and four nuclear genes (c-mos, ACM4, MC1R and RAG2) and their relationships assessed using both phylogenetic and population genetic analyses. The results indicate that both species probably have a vicariant origin and diverged when the Socotra Archipelago had already drifted from mainland (about 17 My ago). Interestingly, the two species present high level of intra-specific genetic variability within Socotra. We discuss the main factors that may have promoted both speciation and intra-island diversification in the genus Haemodracon taking into account the geophysical and climatic events that have shaped the biogeographic history of the Socotra Archipelago.
