Tissue culture derived from apical orthotropic stem segments of Coffea arabica have been established and caffeine production as well as growth parameters were analyzed in function of time. The cultures synthesized relatively high amounts of caffeine compared with generally low in vitro formation of other secondary plant substances. The total content was between 1.0 and 1.6 related to dry weight and corresponds to the caffeine concentration in a coffee bean. We found a positive correlation between callus growth and caffeine formation. In addition we studied the influence of various factors on cafeeine production, i.e. light, phytohormones, media volume and caffeine concentration in the media
