Host-defense peptides (HDP) present many advantages for the treatment of microbial diseases, such as: 1) broad spectrum of activity, 2) unlike classic antibiotics, HDP do not induce resistance; 3) They act synergically with conventional antibiotics; 4) They neutralize endotoxins and are active in animal models. However, it is considered that current natural antimicrobial peptides are not suitable for drug development due to stability and biodisponibility issues. In order to improve their stability, reduce possible toxicity and/or decrease production costs, the production of analogues by chemical modification of “parent” peptides or by biotechnology should be implemented.1 This communication describes the site-selective modification of peptides using different “Customizable Units”,2 derived from serine and threonine, which allow the introduction of a diversity of unnatural chains. The antimicrobial activity of these peptides will be commented as well.
