Elaboration of a new methodology using arbuscular mycorrhizae and rhizobia included in alginate beads for inoculation two species of tropical woody legumes, Acacia senegal and Acacia mangium

The aim of our work was to define a new methodology using arbuscular mycorrhizae and rhizobia included in alginate beads for inoculating Acacia senegal and Acacia mangium, two species of tropical woody legumes. The inocula consisted of liquid medium for rhizobia and leek roots infected by Glomus intraradices. We started by an optimization of the initia protocole in order to include in alginate beads the more infected roots pieces. From these results, it was possible to determine the ratio of parts of infected roots and rhizobia cells which can be included together in alginate beads. Others methods of inoculation were tested and compared with alginate beads containing the both microsymbionts: alginate beads containing infected roots with G. intraradices and coated with a mixture of peat and rhizobia liquid culture; roots plant coated directly with a mixture of alginate / rhizobia liquid culture parts of infected roots with G. intraradices. Our results showed that for A. mangium and A senegal, alginate beads containing the both microsymbionts allow the establishment of al efficient dual symbiosis in plants cultivated in nursery conditions. Regarding the two other methodologies tested, results obtained were similar to those obtained with alginate bead and offer some interesting perspectives in terms of applications. All these results will have to be confirmed through further experiments carried out in nurser conditions and in the field. However, our results suggest that we could identify methodology of inoculation which could be used easily, without sophistication, in nursery fo preparing industrial forestry plantations.
