Picó, Yolanda Ministerio de Economía y Competitividad (España) Póster presentado en el congreso 10th LC/MS/MS Workshop On Environmental Applications And Food Safety, celebrado en Barcelona, España, del 1 al 3 de julio de 2014 The alcohol consumption by population could be estimated using the method based on the measurement of specific urinary metabolites in the influent of the Waste Water Treatment Plant (WWTP). Sewage epidemiology has previously been demonstrated far pharmaceuticals and drugs of abuse (Thomas et al., 2012; Zuccato et al. , 2008). The urinary metabolite that was used for this purpose was the Ethyl sulfate (ES) that is excreted in urine as a minor metabolite (0 .011 % on molar basis) after consume alcoholic beverages, being an useful biomarker (Hoiseth et al., 2008). The aim of this study is to develop a reliable and brief method to determine the ES carried out by ion-pair mass spectrometry. The validated method was applied to sewage samples of the influent of 3 WWTP, Pineda 1, Pineda 11 and Quart-Benager, that treats most of the wastewater of Valencia City and its surrounding towns. Sampling campaign was in 2014 from March 4th to 20th. Two approaches were evaluated. The addition of the ion-pair was into the mobile phase or only to the sample. As well, different ion-pair were tested as tetrabuthylamonium clorure, dihexylamine, tributylamine, acetic acid and ammonium formate. Finally, the samples were prepared by adding 50 mM of the ion-pair in raw waters. Chromatographic separation of ES was performed using an Agilent Technologies 1260 UHPLC. The column was Kinetex C 18 (1.7 ¿m, 100 A, 50 x 2.1 O mm) maintained at temperature of 30 ºC and a constant flow rate of 0.2 ml/min. The mobile phase was eluent A (formic acid 0.1 % in water) and eluent B (formic acid 0.1 % in methanol). The gradient elution was 10% B until 5 min. The injected vol u me was 5 ¿L. The UHPLC was coupled to an Agilent Technologies 641 O triple quadrupole mass spectrometer with an electrospray Turbo V ionization source working in negative ionization (NI) mode, 300ºC gas temperature, 11 L min-1 gas flow and 25 psi nebulizer. The determination of ES in the influent of the selected WWTPs shows that this compound was detected in 100% of influents from Pineda 1, Pineda 11 and Quart-Benager. Its consumption was variable during week and weekend, being higher on weekends.
