Cardiovascular disease and type 2 diabetes are majar causes of death in developed countries. These diseases are associated with each other and linked with oxidative stress. Currently, the research interest has focused on the characterization of food-derived multifunctional peptides with preventive and/or therapeutic potential against chronic disorders. One of the greatest challenges in using bioactive peptides for the modulation of physiological functions is their bioavailability. Survival of gastrointestinal digestion and intestinal epithelial absorption in the human digestive tract are essential prerequisites for bioactive peptides to have any impact. Erythrina edulis (pajuro) is a legume with a broad range of uses because of its nutritional and health promoting properties. This plant contains an elevated content of protein with high quality and digestibility, and potential to release bioactive peptides. The antioxidant properties of pajuro protein hydrolyzates with alcalase have been recently reported. The objective of this research was to identify the peptides contained in those hydrolyzates, and evaluate their multifunctionality and bioavailability. Among 29 peptides identified by MALDI-TOF and HPLC-ESI-MS/MS, 17 were selected according to their structure and synthesized. The antioxidant, anti-inflammatory and dipeptidyl peptidase IV (DPPIV) inhibitory activities were evaluated. Most active fragments were subjected to simulated gastrointestinal digestion and their bioavailability was studied in the Caco-2 cell model. The results confirm the role of pajuro proteins as source of multifunctional and bioavailable peptides with potential as novel ingredients of functional foods to prevent and/or manage diabetes and oxidative-stress related disorders.
